Dear Netters, I am trying to quantify (count) short mycorrhizal and short non-mycorrhizal roots on the root samples of loblolly and slash pine seedlings which I grew in a nursery bed. Half of the seedlings were inoculated with *Pisolithus tinctorius* at planting. The major difficulty which I encounter looking at the root samples under a disecting microscope (magnification 10x to 140x) is that I don't know if monopodial short roots which are club shaped/swollen toward the root tip are or are not mycorrhizal. Does any one know a fast method of identifying ectomycorrhizal roots by STAINING or any other method? Would larger magnification and cross-sectioning help to properly classify the roots in question?
